The Kurachi lab is focusing on molecular mechanisms of CD8+ T cell differentiation and exhaustion.
Memory CD8+ T cells play a central role in mediating protective immunity against intracellular pathogens and cancer. A precise understanding of the mechanisms underlying the formation of memory CD8+ T cells in humans is key for the development of novel vaccines and therapies. In addition, during chronic infections and cancer, CD8+ T cells are exposed to persistent antigenic stimulation. This results in their progressive loss of effector function, a state termed T cell exhaustion. Immunotherapies that blockade immune checkpoint molecules have proven to be very successful against multiple tumor types by reinvigorating exhausted CD8+ T cells. However, only a fraction of patients (approximately 10-30%) benefits from these therapies. Therefore, it is essential to understand the transcriptional and epigenetic regulation of the differentiation path that leads to exhausted CD8+ T cells. The Kurachi lab aim to elucidate molecular mechanisms of CD8+ T cell differentiation by utilizing cutting-edge technologies such as genome editing, next-generation sequencer, and high parameter flowcytometry.
